How does erythromycin affect the metabolism of statins and what risks, such as increased statin toxicity, arise when these medications are co-administered?
Erythromycin inhibits CYP3A4 enzymes, increasing plasma levels of statins metabolized by this pathway, such as simvastatin, raising the risk of myopathy and rhabdomyolysis.
